The lack of effective treatments for non-alcoholic steatohepatitis (NASH) and NASH-associated hepatocellular carcinoma (NASH-HCC) underscores the need to understand the transition from NASH to NASH-HCC. Identifying why some NASH patients develop HCC while others do not is a significant challenge, with growing evidence indicating that immune dysregulation plays a crucial role in this progression. The interstitial space, where cells receive nutrients and signals, significantly impacts immune function, suggesting that understanding the liver"s metabolic environment could reveal how immune cells behave during NASH progression to HCC. By profiling interstitial fluid metabolism and single-cell transcriptomics, the team aims to understand the metabolic regulation of the immune landscape in NASH to NASH-HCC progression. Using a high-fat, high-fructose, and high-cholesterol dietary mouse model that mirrors human progression, the team will study the metabolic and immune changes leading to NASH-HCC. In addition, the team’s preliminary findings show immune alterations and reduced levels of the bile acid hyodeoxycholic acid (HDCA) during NASH-HCC transition. TaRgeted Analysis of Novel Secreted Factors for Obesity Reduction and Metabolic Health (TRANSFORM)

Description:

Multi-omic analysis of secreted factors from various datasets was carried out, including human, mouse and primate plasma and metabolic tissues to identify novel primate-specific secreted factors critically involved in the regulation of physiology balance. Integrative data analysis and in silico rank scoring revealed a list of promising secreted factor candidates. We will be using model organisms (e.g. mice, zebrafish) to conduct phenotypic screens and prioritize candidates for detailed validation, characterization, mode-of-action studies and therapeutic strategies.

Job responsibilities:

● Conduct in vitro wet lab experiments for identified candidates (e.g. cell culture, molecular biology and assays, protein chemistry, pharmacological efficacy etc)

● Conduct in vivo screening experiments for prioritized candidates (e.g. in preclinical models of obesity)

● Conduct end-point and mechanistic experiments (e.g. target tissue and blood collection, sample preparation etc)

● Analyze collected data and maintain accurate monitoring and reporting of experimental set-up, results and research findings

● Interact and collaborate with and assist other researchers and collaborators on related projects

● Communicate regularly with supervisor and join discussions with Principal Investigator and rest of the lab to discuss scientific progress and direction

● Assist in the preparation of grant applications and grant process reports

● Present findings both orally and in written form via presentations and publication in peer-reviewed journal

● Assist in the preparation of figures and manuscripts on the research projects for publication in peer-reviewed journals

● Contribute to other program-related projects and organizations and with the general scientific community across A*STAR

Job Description

Key Responsibilities

• Conduct sensory evaluations, along with chemical and microbiological tests, on both products and raw materials.
• Understand and implement sampling plans, conducting sampling of raw materials and products as required.
• Prepare laboratory culture media and reagents as needed.
• Prepare product samples upon customer request or for internal/external laboratory testing.
• Accurately document all necessary information on the provided inspection forms and master list.
• Maintain cleanliness and organization of the laboratory and its equipment.
• Notify the QC Head of Department (HOD) about lab inventory levels to ensure timely procurement of necessary items and equipment for analysis.
• Report any non-conformities in raw materials and products to the HOD.
• Store retained raw material samples according to production dates for future reference in case of customer quality disputes.
• Participate in company-wide activities and tasks as required.

Role and Responsibilities

Job Requirements:
