As a Scientist, you will be responsible to generate CRISPR/Cas9 mediated edited human embryonic stem cells for ciliary genes, differentiation into airway epithelium, characterisation of ciliary defects, rescue of mutant phenotype with lipid nanoparticle packaged RNA.
Job Responsibilities:
- Use CRISPR/Cas9 gene editing to generate homozygous loss-of-function mutations in DNAI1, CCDC40 and CCDC39 genes in human embryonic pluripotent stem cell line, H9.
- Verify authenticity of the mutants using sequencing of the DNA, cDNA and also using western blotting to ensure loss of the corresponding proteins.
- Differentiate mutant and control cells into airway epithelium and assess the effects on ciliary differentiation and motility using immunofluorescence staining and confocal microscopy as well as video microscopy.
- Examine if lipid nanoparticle encapsulated wild-type RNAs for these genes are able to rescue the ciliary defects using immunofluorescence staining and confocal microscopy as well as video microscopy.
Job Requirements:
- PhD in Stem Cells/ Developmental Biology/ Genetics or other related disciplines.
- Strong research background in embryonic stem cell culture and manipulation, confocal microscopy.
- Proficiency in stem cell culture and gene editing in stem cells using CRISPR/Cas9.
