Human kinases and epigenetic proteins are important therapeutic targets. Recent development of selective and potent inhibitors of proteins has yielded remarkable results in disease treatment, with numerous drugs already on the market, and many more undergoing clinical trials. Notwithstanding, the discovery of new covalent kinase and epigenetic drugs remains highly challenging, including issues related to selectivity, efficacy and acquired drug resistance. Taking cue from the kinase drug discovery field, the hypothesis of these projects is that a universal strategy for targeted covalent inhibitors (TCIs) capable of potent target inhibition and minimal acquired drug resistance is possible, if highly effective small molecules could be developed to covalently target non-cysteine (C) nucleophilic residues within/near the active-site of kinase/epigenetic proteins. Our recent discovery of covalent kinase inhibitors against BCR-ABL (a key kinase for CML) in which CBA/SA and EBA warheads formed stable, (reversible and irreversible) covalent linkage with catalytic lysine (K) in Abl kinase under cellular environments indicates other covalent chemistries are feasible. Therefore, the goal of the current proposals is to confirm covalent chemistries targeting other nucleophilic residues (E/D/R/M/etc) are possible, and amenable to the development of cell-active, selective TCIs for various therapeutic proteins.

The scope of this proposal will be limited to the fundamental basic science, and NOT the actual clinical implementation, of the developed chemical/chemical biology tools. The Mechanobiology Institute, Singapore, is a multidisciplinary institute committed to developing new paradigms for biomedical research by focusing on the quantitative analysis of dynamic functional processes. Asst. Prof. Chan seeks a highly motivated individual to join his laboratory on Mammalian Development and Tissue Hydraulics as a postdoctoral fellow at the Mechanobiology Institute in the National University of Singapore. This position is offered under a highly interdisciplinary project that aims to understand macrophage-environment mechanochemical crosstalk during ovarian ageing.

Project Scope

The candidate will investigate the mechanobiological principles by which cell mechanics, ECM remodelling and inflammation drive fibrosis during ovarian ageing. The selected candidate will combine an ex vivo approach with advanced imaging, omics and biophysics to explore how tissue-resident macrophages respond to age-associated changes in matrix stiffness and composition, and how this in turn drives macrophage-mediated fibrosis. The Earth Observatory of Singapore (EOS) at Nanyang technological University in Singapore invites applications for the position of Research Assistant to support the SouthEast Asia SEA-level (SEA2) Program and the Climate Transformation Programme (CTP).These are interdisciplinary research programmes that integrate sea-level and climate datasets in Southeast Asia with sophisticated modelling capabilities to improve the accuracy of projections of sea-level rise and climate change.We are looking for someone with a passion for making Singapore and Southeast Asia a safer and more sustainable place in the face of climate change, sea-level rise, and their associate hazards, through supporting and communicating critical research on sea-level science.

The ideal candidate will have experience in both field work and laboratory work. The primary responsibilities include managing the laboratory, ensuring health and safety (for both the laboratory and its users), and organizing field work. Ideally, they will have a working knowledge of machines used for biogeochemical analyses, such as ICP, iSEAL, CHN analysers, and GCMS. They will also assist in research support and administrative duties (i.e., booking of cars, doing purchase orders and goods receipts, as well as other relevant duties). The incumbent will also work closely with other EOS Researchers on their projects, laboratory work, and field work.

The successful candidate will have strong planning and organizational skills, ability to provide guidance to students, and work both independently and in a team. They must possess a keen interest in Earth and/or Environmental sciences, sea-level rise, and climate change. Experience in driving and laboratory management is highly desirable.

The candidate will examine the conduct high-resolution ocean model (ROMS) simulations to study past and future marine heatwave events in Singapore and examine their impacts on marine ecosystems. The position is a key component of the Marine Climate Chance Science (MCCS) Programme aimed at assessing the long-term viability of mangroves and coral reefs as nature-based climate solutions in Singapore. The Research Fellow will work in a multidisciplinary research team, including coral reef geomorphology, sea level studies, climate modelling, and sedimentology to understand the response of Singapore’s coastal and marine ecosystems to anthropogenic and climate change impacts.

Key Responsibilities:

• Set up and run high-resolution ocean model (ROMS) simulations to study past and future marine heatwaves in Singapore (and the wider SE Asia region)
• Global climate model (e.g., CMIP6) data analysis related to the project goals
