Understanding the pharmaco/ nutrico/ toxicokinetic behaviour of drugs, chemicals and food ingredients is a fundamental step in assessing the efficacy and safety of these materials. It can be challenging to study such substances directly in human subjects, while animal models lack direct translatability to humans due to inherent differences in species biology. One promising strategy is the use of physiologically-based pharmacokinetic (PBPK) modelling to predict the local tissue and systemic exposure of these substances. We have developed in-house in vitro and in silico tools to model chemical absorption, distribution, metabolism and excretion (ADME) in humans using data generated from in vitro assays. We apply these tools to predict pharmacokinetic profiles in different settings, ranging from oral or transdermal administration to studying special populations such as pregnant women, for a variety of substances including pharmaceuticals, food ingredients, environmental contaminants, essential micronutrients and microbial metabolites. These capabilities are further enhanced with state-of-the-art, enabling technologies such as the SHIME® intestinal digestion simulator and spatial mass spectrometry imaging.

The candidate in this position will be expected to be involved in the following activities:

• Literature search to extract relevant ADME data for chemicals of interest.
• Perform in vitro absorption, transporter, metabolism, fraction unbound and other related ADME assays.
• Quantify membrane protein, metabolic enzyme and xenobiotic concentrations within biological samples using liquid chromatography-tandem mass spectrometry (LC/MS/MS) instruments.
• Integrate appropriate in vitro and in silico data into PBPK models to predict the ADME of chemicals.

Our team aims to leverage on stem cell biology (Sagi, Chia et al., Nature 2016; Chia et al., Nature Cell Biology 2019; Chia et al., Nature Cell Biology 2017) and cell engineering to develop alternative immunotherapeutic strategies to target refractory cancer types. The candidate will work closely with clinicians to establish empirical platforms for identifying and validating cancer vaccine candidates, and to translate research findings into clinical practice.

Also, the candidate will be involved in the assessment and development of vaccines against novel antigens for the treatment of cancer and other human diseases. We are seeking applications for two full-time postdoctoral researcher/research fellow positions to work on building a theoretical framework to examine factors that affect public trust in digitalization, alongside trust in government and public institutions to manage associated risks. Through multi-city large scale surveys and online experiments, the study seeks to understand public attitudes towards technology, the digital information pipeline, and the role of government. This research will contribute to policy design and the implementation of digital technologies in a way that is sensitive to public concerns and expectations.

The two positions are available from Q3 2024 (reasonably earlier or later start date is negotiable) for 3 years each. The researchers are expected to have significant experience in designing and conducting survey research with a focus on digital technologies, platforms, AI, public trust, or other closely related areas. The project using a policy design lens will contribute to the literature on building trust in digitalization through mixed-method research in multiple phases. The project will conceptualise trust in digital information and examining factors such as trust in technology and trust in government.
