Job Description

The Tan lab is looking for a research assistant who will work with Assistant Professor Tan Yong Zi on biochemistry and structural biology under a project entitled Understanding the Structure and Function of Mammalian Membrane Proteins Involved in Human Diseases.

While membrane proteins only make up around 20% of the proteome, they constitute around 60% of drug targets. However, high resolution structural information is severely lacking for them, with only about 2% of the structures deposited in the public protein database (PDB) being annotated as membrane proteins. The Tan lab (www.tyzlab.com) strives to fill in this gap by targeting membrane proteins involved in human diseases such as cancer and diabetes. Our forte is using state-of-the-art single-particle cryogenic electron microscopy (cryo-EM) to obtain high-resolution atomic structures of these membrane proteins. We also actively work on methods development in cryo-EM that will help us answer our biological questions.

The Tan lab is generously funded by NRF Fellowship, MOE and NUS Presidential Young Professorship under the Department of Biological Sciences, NUS. We are a core member of the Centre for BioImaging Sciences' (CBIS), and have access to high-end cryo-EM microscopes, including a Titan Krios with K3 direct detector and energy filter.

We are seeking a highly motivated Research Fellow to join our research team in the Materials Science/Physics Department at the National University of Singapore. The candidate will work on fabricating and characterizing twisted bilayer graphene (tBLG) using large-scale CVD techniques, aiming to revolutionize the production of high-quality, defect-free graphene films for advanced optoelectronic devices like single-photon detectors.

The Mechanobiology Institute (MBI) at the National University of Singapore (NUS) is a leading multi­disciplinary institute devoted to developing new paradigms for understanding biological functions in health and diseases from the perspective of cell, tissue and organ mechanics/dynamics and mechano-signal transduction. Understanding the pharmaco/ nutrico/ toxicokinetic behaviour of drugs, chemicals and food ingredients is a fundamental step in assessing the efficacy and safety of these materials. It can be challenging to study such substances directly in human subjects, while animal models lack direct translatability to humans due to inherent differences in species biology. One promising strategy is the use of physiologically-based pharmacokinetic (PBPK) modelling to predict the local tissue and systemic exposure of these substances. We have developed in-house in vitro and in silico tools to model chemical absorption, distribution, metabolism and excretion (ADME) in humans using data generated from in vitro assays. We apply these tools to predict pharmacokinetic profiles in different settings, ranging from oral or transdermal administration to studying special populations such as pregnant women, for a variety of substances including pharmaceuticals, food ingredients, environmental contaminants, essential micronutrients and microbial metabolites. These capabilities are further enhanced with state-of-the-art, enabling technologies such as the SHIME® intestinal digestion simulator and spatial mass spectrometry imaging.

The candidate in this position will be expected to be involved in the following activities:

• Literature search to extract relevant ADME data for chemicals of interest.
• Perform in vitro absorption, transporter, metabolism, fraction unbound and other related ADME assays.
• Quantify membrane protein, metabolic enzyme and xenobiotic concentrations within biological samples using liquid chromatography-tandem mass spectrometry (LC/MS/MS) instruments.
• Integrate appropriate in vitro and in silico data into PBPK models to predict the ADME of chemicals.
