A Research fellow position is available in Dr. Anand Andiappan Laboratory at the Singapore Immunology Network (SIgN). The successful candidate will support the research work that focuses on studying inflammaeing in the context of skin aging in Asian populations.

The candidate will work on primary cells from cohorts and cell culture systems towards understanding functional immune responses in skin immune interactions and recapitulating them in in-vitro and ex-vivo systems. Additional work involves cellular and stimulation assays with flow cytometry and FACS analysis along with imaging by microscopy, and associated data analysis with statistical tested.

TaRgeted Analysis of Novel Secreted Factors for Obesity Reduction and Metabolic Health (TRANSFORM)

Description:

Multi-omic analysis of secreted factors from various datasets was carried out, including human, mouse and primate plasma and metabolic tissues to identify novel primate-specific secreted factors critically involved in the regulation of physiology balance. Integrative data analysis and in silico rank scoring revealed a list of promising secreted factor candidates. We will be using model organisms (e.g. mice, zebrafish) to conduct phenotypic screens and prioritize candidates for detailed validation, characterization, mode-of-action studies and therapeutic strategies.

Job responsibilities:

The funded project will aim to characterize and compare host-alphavirus interactions at the immune response level in vivo in animal models through a novel viral-centric approach. Preliminary data of the lab has shown a differential adaptive immune response polarization between two alphaviruses and identified, via in vivo comparative multiOmic approaches, a set of candidate host factors potentially responsible for the phenotype. The project will entail a close characterization of these host factors and their role in the viral life cycle as well as the modulation of the immune response of small animal model to these viruses. In addition to multi-Omics approaches (including single-cell and spatial), protein-protein interactions, microscopy, functional genomics, viral reverse genetics and CRISPR Cas9 gene editing will be some of the tools used to investigate this viral hijacking and understand the host-viral interplay. Animals models available in the lab include mice and zebrafish. Downstream of characterization will aim to validate these targets in small preclinical models for subsequent drug discovery in the A*STAR ecosystem. Responsibilities

The lab of RNA in Disease and Translational Technologies at the Institute of Molecular and Cell Biology, A*STAR, Singapore is seeking to recruit a highly motivated Scientist or Senior Scientist to join a team developing a ribozyme-based RNA platform for potential translational and/or clinical applications.

The work will involve both in vitro and in vivo techniques, including molecular biology (RNA design, in vitro transcription, cell-free RNA assays, RT-qPCR, molecular cloning), cell biology (cell culture, cell transfection, flow cytometry, antisense oligonucleotides and RNAi, CRISPR), high-resolution imaging techniques, and experiments in mouse models (e.g. transgenesis, in situ hybridization and immunohistochemistry, imaging and functional analyses)

The candidate will work on primary cells from cohorts and cell culture systems towards understanding functional immune responses in skin immune interactions and recapitulating them in in-vitro and ex-vivo systems. Additional work involves cellular and stimulation assays with flow cytometry and FACS analysis along with imaging by microscopy, and associated data analysis with statistical tested.
