Description:

The lack of effective treatments for non-alcoholic steatohepatitis (NASH) and NASH-associated hepatocellular carcinoma (NASH-HCC) underscores the need to understand the transition from NASH to NASH-HCC. Identifying why some NASH patients develop HCC while others do not is a significant challenge, with growing evidence indicating that immune dysregulation plays a crucial role in this progression. The interstitial space, where cells receive nutrients and signals, significantly impacts immune function, suggesting that understanding the liver"s metabolic environment could reveal how immune cells behave during NASH progression to HCC. By profiling interstitial fluid metabolism and single-cell transcriptomics, the team aims to understand the metabolic regulation of the immune landscape in NASH to NASH-HCC progression. Using a high-fat, high-fructose, and high-cholesterol dietary mouse model that mirrors human progression, the team will study the metabolic and immune changes leading to NASH-HCC. In addition, the team’s preliminary findings show immune alterations and reduced levels of the bile acid hyodeoxycholic acid (HDCA) during NASH-HCC transition. The identified candidate will also test the potential of HDCA in preventing NASH to HCC transition, and further explore HDCA"s effects on immune cells.

DHA supports innovators in driving adoption and scaling of market ready or near-market ready digital health solutions. DHA is hosted by Singapore's Diagnostics Development Hub (DxD Hub). The DHA integration engineer is responsible for planning and developing workflow and technical interfaces between healthcare IT systems. She/He will work with DHA Project Managers, Solution Architects, health systems and companies to accomplish this objective. She/He may also be called upon to advise other developers on integration standards.

Responsibilities:

• You’ll be involved in the development, design, build, testing, and go live phases of multiple integration projects.
• This includes scripting to conform / transform to another systems specifications.

TaRgeted Analysis of Novel Secreted Factors for Obesity Reduction and Metabolic Health (TRANSFORM)

Description:

Multi-omic analysis of secreted factors from various datasets was carried out, including human, mouse and primate plasma and metabolic tissues to identify novel primate-specific secreted factors critically involved in the regulation of physiology balance. Integrative data analysis and in silico rank scoring revealed a list of promising secreted factor candidates. We will be using model organisms (e.g. mice, zebrafish) to conduct phenotypic screens and prioritize candidates for detailed validation, characterization, mode-of-action studies and therapeutic strategies.

Job responsibilities:

● Conduct in vitro wet lab experiments for identified candidates (e.g. cell culture, molecular biology and assays, protein chemistry, pharmacological efficacy etc)

● Conduct in vivo screening experiments for prioritized candidates (e.g. in preclinical models of obesity)

● Conduct end-point and mechanistic experiments (e.g. target tissue and blood collection, sample preparation etc)

Responsibilities

The lab of RNA in Disease and Translational Technologies at the Institute of Molecular and Cell Biology, A*STAR, Singapore is seeking to recruit a highly motivated Scientist or Senior Scientist to join a team developing a ribozyme-based RNA platform for potential translational and/or clinical applications.

The work will involve both in vitro and in vivo techniques, including molecular biology (RNA design, in vitro transcription, cell-free RNA assays, RT-qPCR, molecular cloning), cell biology (cell culture, cell transfection, flow cytometry, antisense oligonucleotides and RNAi, CRISPR), high-resolution imaging techniques, and experiments in mouse models (e.g. transgenesis, in situ hybridization and immunohistochemistry, imaging and functional analyses)

A Research fellow position is available in Dr. Anand Andiappan Laboratory at the Singapore Immunology Network (SIgN). The successful candidate will support the research work that focuses on studying inflammaeing in the context of skin aging in Asian populations.

The candidate will work on primary cells from cohorts and cell culture systems towards understanding functional immune responses in skin immune interactions and recapitulating them in in-vitro and ex-vivo systems. Additional work involves cellular and stimulation assays with flow cytometry and FACS analysis along with imaging by microscopy, and associated data analysis with statistical tested.

Duties will include managing safety in the laboratory, creating Standard Operating Procedures (SOPs) and laboratory risk assessments, maintaining laboratory inventories, perform other laboratory administrative duties and operations such as ordering and tracking laboratory budget.

We are looking for talented wet-lab Research Fellow to work with a multidisciplinary group of scientists on the discovery of new targets for therapy in Diabetic Nephropathy (DN), a devastating complication of diabetes causing significant morbidity, mortality and human suffering. In Singapore, the incidence of DN as a cause of end-stage kidney disease is among the highest in the world. The job will be part of the NMRC-funded project called Diabetes Study in Nephropathy and other Microvascular Complications II, and it will be based within the Centre for Computational Biology (CCB).
