Description:

Multi-omic analysis of secreted factors from various datasets was carried out, including human, mouse and primate plasma and metabolic tissues to identify novel primate-specific secreted factors critically involved in the regulation of physiology balance. Integrative data analysis and in silico rank scoring revealed a list of promising secreted factor candidates. We will be using model organisms (e.g. mice, zebrafish) to conduct phenotypic screens and prioritize candidates for detailed validation, characterization, mode-of-action studies and therapeutic strategies.

Job responsibilities:

The project will entail a close characterization of these host factors and their role in the viral life cycle as well as the modulation of the immune response of small animal model to these viruses. In addition to multi-Omics approaches (including single-cell and spatial), protein-protein interactions, microscopy, functional genomics, viral reverse genetics and CRISPR Cas9 gene editing will be some of the tools used to investigate this viral hijacking and understand the host-viral interplay. Downstream of characterization will aim to validate these targets in small preclinical models for subsequent drug discovery in the A*STAR ecosystem. The program has a strong track record of publication in top-tier journals, successful commercialisation efforts leading to spin-offs, as well as nurturing early career researchers into independent academic or industry positions.

Metabolic dysfunction-associated steatohepatitis (MASH) is a severe metabolic liver disease that can affect up to 6.5% of the adult population, with only one FDA-approved drug. Dr. Jin Zhou’s team is specialised in studying the novel molecular pathogenesis underlying MASH for the identification of new druggable targets. The lab uses a combination of biochemistry, genetics, animal modeling, and metabolomics approaches to understand cellular processes related to mitochondrial function and hepatic lipid metabolism.
