Job Description

The successful candidate will work with Prof YAO Shao Qin on chemical biology/drug discovery-related research under a project on "Targeted covalent Inhibitors against key epigenetic proteins (NMRC)".

Human kinases and epigenetic proteins are important therapeutic targets. Recent development of selective and potent inhibitors of proteins has yielded remarkable results in disease treatment, with numerous drugs already on the market, and many more undergoing clinical trials. Notwithstanding, the discovery of new covalent kinase and epigenetic drugs remains highly challenging, including issues related to selectivity, efficacy and acquired drug resistance. Taking cue from the kinase drug discovery field, the hypothesis of these projects is that a universal strategy for targeted covalent inhibitors (TCIs) capable of potent target inhibition and minimal acquired drug resistance is possible, if highly effective small molecules could be developed to covalently target non-cysteine (C) nucleophilic residues within/near the active-site of kinase/epigenetic proteins. Our recent discovery of covalent kinase inhibitors against BCR-ABL (a key kinase for CML) in which CBA/SA and EBA warheads formed stable, (reversible and irreversible) covalent linkage with catalytic lysine (K) in Abl kinase under cellular environments indicates other covalent chemistries are feasible. Therefore, the goal of the current proposals is to confirm covalent chemistries targeting other nucleophilic residues (E/D/R/M/etc) are possible, and amenable to the development of cell-active, selective TCIs for various therapeutic proteins.

The scope of this proposal will be limited to the fundamental basic science, and NOT the actual clinical implementation, of the developed chemical/chemical biology tools. Therefore, only cell-based and basic animal models will be tested on newly developed, select small-molecule TCIs. Experience/skills required are organic synthesis/chemical biology/protein chemistry/proteomics/molecular biology/cell biology (only some of above skills are required)

We are looking for a highly motivated research officer who is interested in developing next-generation DNA and RNA therapeutics for treating genetic diseases. The individual will be involved in a multi-disciplinary project to develop new technologies and tools for precision editing of nucleic acids. There will be opportunities to further develop techniques in protein engineering, directed evolution, and synthetic biology.

The successful candidate will work with Dr Gloryn Chia on developing cancer vaccines.

We are seeking a highly motivated Research Fellow with expertise in immunology. Our team aims to leverage on stem cell biology (Sagi, Chia et al., Nature 2016; Chia et al., Nature Cell Biology 2019; Chia et al., Nature Cell Biology 2017) and cell engineering to develop alternative immunotherapeutic strategies to target refractory cancer types. The candidate will work closely with clinicians to establish empirical platforms for identifying and validating cancer vaccine candidates, and to translate research findings into clinical practice. Also, the candidate will be involved in the assessment and development of vaccines against novel antigens for the treatment of cancer and other human diseases. The successful candidate will need to independently design and execute experiments, troubleshoot issues, interpret results and give project updates. The Mechanobiology Institute, Singapore, is a multidisciplinary institute committed to developing new paradigms for biomedical research by focusing on the quantitative analysis of dynamic functional processes. Through quantitative biophysical approaches, we address major problems in biomedical sciences, including pathogenesis (from bacterial to cancer), tissue regeneration, and function.

Asst. Prof. Chan seeks a highly motivated individual to join his laboratory on Mammalian Development and Tissue Hydraulics as a postdoctoral fellow at the Mechanobiology Institute in the National University of Singapore. This position is offered under a highly interdisciplinary project that aims to understand macrophage-environment mechanochemical crosstalk during ovarian ageing.

Project Scope

The candidate will investigate the mechanobiological principles by which cell mechanics, ECM remodelling and inflammation drive fibrosis during ovarian ageing. The selected candidate will combine an ex vivo approach with advanced imaging, omics and biophysics to explore how tissue-resident macrophages respond to age-associated changes in matrix stiffness and composition, and how this in turn drives macrophage-mediated fibrosis.
