The successful candidate will work with Prof YAO Shao Qin on chemical biology/drug discovery-related research under a project on "Targeted covalent Inhibitors against key epigenetic proteins (NMRC)".

Human kinases and epigenetic proteins are important therapeutic targets. Recent development of selective and potent inhibitors of proteins has yielded remarkable results in disease treatment, with numerous drugs already on the market, and many more undergoing clinical trials. Notwithstanding, the discovery of new covalent kinase and epigenetic drugs remains highly challenging, including issues related to selectivity, efficacy and acquired drug resistance. Taking cue from the kinase drug discovery field, the hypothesis of these projects is that a universal strategy for targeted covalent inhibitors (TCIs) capable of potent target inhibition and minimal acquired drug resistance is possible, if highly effective small molecules could be developed to covalently target non-cysteine (C) nucleophilic residues within/near the active-site of kinase/epigenetic proteins. Our recent discovery of covalent kinase inhibitors against BCR-ABL (a key kinase for CML) in which CBA/SA and EBA warheads formed stable, (reversible and irreversible) covalent linkage with catalytic lysine (K) in Abl kinase under cellular environments indicates other covalent chemistries are feasible. Therefore, the goal of the current proposals is to confirm covalent chemistries targeting other nucleophilic residues (E/D/R/M/etc) are possible, and amenable to the development of cell-active, selective TCIs for various therapeutic proteins.

The scope of this proposal will be limited to the fundamental basic science, and NOT the actual clinical implementation, of the developed chemical/chemical biology tools. Therefore, only cell-based and basic animal models will be tested on newly developed, select small-molecule TCIs. We are looking for a highly motivated research officer who is interested in developing next-generation DNA and RNA therapeutics for treating genetic diseases. The individual will be involved in a multi-disciplinary project to develop new technologies and tools for precision editing of nucleic acids. There will be opportunities to further develop techniques in protein engineering, directed evolution, and synthetic biology.

Responsibilities:

• Design and clone constructs for experiments
• Perform activity characterization in vitro and in mammalian cells
• Perform qPCR/RT-qPCR of mammalian cells
• Perform high-throughput sequencing library preparation
• Analyze data and communicate results of experiments to colleagues
• Maintenance of laboratory equipment and general laboratory supplies

Job Requirement

• Bachelor’s Degree in Biochemistry, Biotechnology, Bioengineering, Biological Sciences, Chemistry, Chemical Engineering, or other related fields
• Experience with mammalian cell culture and molecular biology techniques (eg. molecular cloning, PCR, gel electrophoresis, miniprep)
• Experience with flow cytometry, protein expression, bacteriophage, high-throughput sequencing, or bioinformatics will be valued
• Excellent organizational and interpersonal skills
• Highly motivated, passion for science, committed to laboratory work
• Ability to work independently and as part of a team
• Excellent communication (verbal and written) and presentation skills
• Curiosity to learn new knowledge and skills

• Support in the monthly stock take of finished goods.
• Process delivery orders and inventory update.

Requirements:

Job Description

The successful candidate will work with Dr Gloryn Chia on developing cancer vaccines.

We are seeking a highly motivated Research Fellow with expertise in immunology. Our team aims to leverage on stem cell biology (Sagi, Chia et al., Nature 2016; Chia et al., Nature Cell Biology 2019; Chia et al., Nature Cell Biology 2017) and cell engineering to develop alternative immunotherapeutic strategies to target refractory cancer types. The candidate will work closely with clinicians to establish empirical platforms for identifying and validating cancer vaccine candidates, and to translate research findings into clinical practice. Also, the candidate will be involved in the assessment and development of vaccines against novel antigens for the treatment of cancer and other human diseases. The successful candidate will need to independently design and execute experiments, troubleshoot issues, interpret results and give project updates. Main Purpose of Job:

• The Regional Research & Development (R&D) Manager will be primarily responsible to oversee and manage the full spectrum of R&D Division activities in Yanmar International Singapore Pte Ltd (YIS).
• To establish a close partnership with regional R&D teams in the designated overseas companies and with Japan headquarters (HQ) R&D Senior Management to ensure contribution of regional R&D initiatives.

Responsibilities:

• Develop the regional R&D strategy plan as part of HQ’s R&D strategy planning process and provide input on business priorities and challenges faced regionally.
• To work with the HQ R&D Division and regional Corporate Planning departments to ensure that R&D programs formulated supports the business needs, and act as a change advocate to help drive these initiatives into each business.
• Determine regional short-term and mid-term R&D roadmap and strategies in accordance with the HQ’s R&D strategies and business requirements.
• Oversee all research, planning, and implementation of new programs and protocols for the designated overseas companies including the development of new products.
• Coordinate with overseas R&D departments on the implementation of activities related to Product Development and Product Testing areas, as well as for the improvement of existing products and technologies.

• Fluent in both spoken and written English. Ability to speak and understand Japanese language is an added advantage.
