Process Development Joint Lab (PDJL) between Bioprocessing Technology Institute (BTI), Agency for Science, Technology and Research (A*STAR) and Advanced Cell Therapy and Research Institute Singapore (ACTRIS) aims to advance pre-GMP process and analytical development of cell therapy assets to support seamless transition towards GMP manufacturing. The PDJL will work closely with industry solution providers and in partnership with ACTRIS collaboratively support and advance development of novel cell therapy assets from local academia, national healthcare groups and biotech companies with the ultimate aim of translating them into GMP-compliant workflows for clinical-scale manufacturing.

We are looking for experienced Analytical Development Scientist with GMP, GLP and CMC experience to join this exciting translational fronting initiative to support with Process and Analytical Development for advancing cell therapy modalities. Selected candidates will support with project management, technology transfer activities and will be actively involved in planning and execution of analytical development studies in order to establish fit-for-purpose release assay methods for tech transfer to GMP manufacturing. Roles and responsibilities of candidates will include but not limited to the following:

· Technology Transfer of cell therapy pipelines (CAR-T, TCR-T, NK, ??-T, TILs, iPSC-derived cells) from asset developers to PDJL and subsequent transfer of established methods and SOPs to GMP manufacturing team

· Develop and optimize standardized assay methods for evaluating critical quality attributes of manufactured cells.

o Develop and optimize multi-parameter flow cytometry for phenotyping and characterizing cell populations

o Develop and optimize vector copy number determination using droplet digital PCR or quantitative real-time PCR.

o Develop and optimize functional and potency measurement of cells such as multiplex cytokine secretion assay, ELISA assay and cytotoxic cell killing assay.

o Support with further characterization of cells where required (serial killing assay, proteomic, transcriptomic, metabolomic analysis)

· Support with process related investigations and root cause analysis for issues arising during GMP manufacturing runs at CMO and support with necessary follow-up studies.

· Support Project management activities to ensure efficient and timely execution of activities, meticulous documentation of studies in GLP compliant manner and routine communication to update GMP partners and CT asset owners

· Perform data analysis, prepare technical reports, establish standard assay methodology for tech transfer to GMP team

· Training of GMP personal or QC personal on analytical assays

· Provide scientific guidance to Associate Scientists

· Perform other related duties incidental to the work described therein.

Job Requirements

· Masters in Immunology, LifeSciences, Biomanufacturing or in a relevant Science field or equivalent BioEngineering or Biotechnology field.

Research overview

Fluorescent labels are essential tools in many branches of life science research. They provide high sensitivity for detection and are non-destructive to the target samples. Traditionally, fluorescent labeling is achieved by adding a chromophore to biomolecules such as proteins or nucleic acids. Conjugated oligoelectrolytes (COEs) are emerging as a new class of fluorescent molecules used to label cells and other specimens for non-invasive fluorescence imaging directly.

COEs are defined by a linear hydrophobic conjugated backbone with polar ionic pendants at both terminals. They were firstly synthesized in Prof. Bazan’s group as two-photo absorption chromophores and later found to have biological applications. COEs are membrane intercalating molecules that span the lipid bilayer. The spontaneous intercalation into membranes from aqueous media results in significant increases in fluorescence emission. By tuning the molecular framework in the conjugated core of COE, COEs characterized by different emission wavelengths can be achieved, thereby gaining access to a range of different colours for fluorescent labelling.

About the Company and the role

Acoerela Pte. Ltd. is a spin off from NUS that is commercializing COEs for use as dyes in life science. This role aims to determine the labelling efficiency of various COE dyes across different biological samples, including but not limited to mammalian cells, bacteria of different membrane structures, and extracellular vesicles. The Metta Home for the Disabled and Metta Home Day Activity Centre were set up in December 2000 to offer community care options to adults with intellectual disabilities who are aged between 18 and 55.

Funded by the Ministry of Community Development, Youth and Sports (MCYS), the former is a purpose-built facility designed to accommodate 103 residents, whereas the latter is supported under the MSF and Tote Board Social Service Fund to provide day care services for a capacity of 50 beneficiaries. Our programmes aim to equip all recipients with the skills and means to lead independent and fulfilling lives, so as to achieve social integration and quality of life.

Tending them with holistic care and working closely with their primary caregivers to provide well-rounded support is a multi-disciplinary team comprising a social worker, training officers, staff nurses, assistant nurses, nursing and therapy aides.

As a Training Officer, the candidate is responsible in the following areas:

1. Supporting of clients

• Cares for and ensures clients’ hygiene and cleanliness
• Support clients in the maintenance/development of daily living skills and community living skills
• Support and trains clients in the maintenance/development of acceptable social and behavioural skills
• Cooperate with caregivers in supporting client’s development and progress
• Work as part of multi-disciplinary team to implement individual training/care plan for clients
• Recommend and implement new programmes for client’s benefit

2. Others

Job Responsibilities

• Execute service contracts from research to clinical grade EVs/Exosomes for upstream and downstream bioprocessing.
• Utilize Tide Motion to generate EVs/Exosomes alongside with characterization via FACS and equivalent techniques.
• Perform process characterization as needed based on project scope.
• Work jointly with other departments (DSP processing/analytical development/QC) to develop end to end continuous process with process intensification.
• Be involved with cross-functional team for Internal transfer from GLP into clinical trial manufacturing which may include validation (e.g. FAT) and engineering runs.
• Write and execute experimental plans as well as white papers, technical reports, presentations.
